What is 3-HO-PCP?

3-HO-PCP was first synthesized in 1978 to study the structure-activity relationship of phencyclidine (PCP) derivatives. In the 1980s, it was further investigated along with PCP, where it was discovered to possess μ-opioid agonist activity in animal models.
Like other substances in its class, notably methoxetamine (MXE), phencyclidine (PCP) and 3-MeO-PCE, it is known to primarily induce a state known as ‘dissociative anaesthesia’, although the extent to which this occurs has not been reported to be highly dose-dependent and variable. It is not known whether the opioid properties studied in animal models are applicable to humans. However, some early reports suggest that this is not the case.
Today, 3-HO-PCP is sold almost exclusively as a research chemical in the gray area by online retailers. Very little is known about its pharmacology, metabolism, and toxicity in humans. Chemical properties
3-HO-PCP or 3-hydroxyphencyclidine is a synthetic dissociative from the class of arylcyclohexylamines. The structure of 3-HO-PCP consists of cyclohexane, a six-membered saturated ring, attached to R1 by two additional rings. One of these rings is a piperidine ring, a nitrogen-containing six-membered ring attached to its nitrogen group. The other ring is an aromatic phenyl ring substituted with a hydroxy group at R3.
3-HO-PCP is a structural analogue of PCP and a homologue 3-MeO-PCP.
Pharmacological effects
Like other arylcyclohexylamine dissociatives, 3-HO-PCP acts primarily as an NMDA receptor antagonist. The NMDA receptor (N-methyl-D-aspartate), a specific subtype of the glutamate receptor, modulates the transmission of electrical signals between neurons in the brain and spinal cord. In order for the signals to pass, the receptor must be open. Dissociatives inhibit the normally functioning NMDA receptors by binding to and blocking them. This disruption of neural network activity, some research suggests, leads to network disintegration through hyperconnectivity throughout the brain. This leads to an increase in noise (random, nonsense and erroneous data) in the brain network and thus leads to the loss of normal cognitive and affective processing, psychomotor function and anesthesia. This is often observed in individuals exhibiting psychosis or induced in healthy participants by high doses of intravenous THC or ketamine.
In contrast to many other dissociatives, 3-HO-PCP has also been found to have significant affinity as a μ-opioid receptor agonist in animal models. However, the extent to which this applies to humans is not yet known.
